目的 探讨体外连续培养过程中人恒牙牙髓细胞向成牙本质样细胞分化的可行性。方法 采用细胞连续培养、酶动力学法、透射电镜观察、免疫组化等方法 ,对体外人牙髓细胞在连续培养过程中的生物学特性进行研究。结果 部分人牙髓细胞在体外可以出现复层生长 ,形成细胞结节 ,并可进一步钙化 ;与成牙本质细胞相似 ,它们具有高碱性磷酸酶活性 ,可合成Ⅰ型胶原 ,其超微结构有许多与成牙本质细胞相似的特征 ,胞间基质中可见膜被基质小泡 ,某些基质小泡内含针状晶体。结论 体外连续培养的人牙髓细胞可向成牙本质细胞分化 ,此过程可作为研究体内牙髓损伤修复细胞增殖、分化的体外模型
Objective To investigate the feasibility of differentiation of human dental pulp cells into odontoblast-like cells during in vitro continuous culture. Methods The biological characteristics of human dental pulp cells during continuous culture were studied by continuous culture, enzyme kinetics, transmission electron microscopy and immunohistochemistry. Results Some human dental pulp cells could grow in vitro in vitro, forming nodules of cells and further calcification. Similar to odontoblasts, they had high alkaline phosphatase activity and could synthesize type I collagen. Its ultrastructure There are many features similar to odontoblasts, the membrane is seen in the interstitial matrix matrix vesicles, some matrix vesicles containing acicular crystals. Conclusion Human dental pulp cells cultured in vitro can differentiate into odontoblasts. This process can be used as an in vitro model to study the proliferation and differentiation of dental pulp cells in vivo
