目的探讨碱性成纤维细胞生长因子(fibroblast growth factor,FGF)对人肝癌细胞(BEL-7402)蛋白激酶B(protein kinase B,PKB)活性的影响。方法以碱性FGF(b FGF)和wortmannin处理人肝癌细胞(BEL-7402),Western blot检测细胞外信号调节蛋白激酶B的活性。结果 25 ng/ml bFGF刺激人肝癌细胞(BEL-7402)10 min,细胞膜和胞质PKB酶活性达高峰。Wortmannin预处理后,人肝癌细胞(Bel-7402)细胞膜和胞质PKB活性在各时间点均明显降低。结论 bFGF能通过磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3K)途径迅速激活人肝癌细胞(BEL-7402)的PKB活性。
Objective To investigate the effect of basic fibroblast growth factor (FGF) on the activity of protein kinase B (PKB) in human hepatoma cell line (BEL-7402). Methods Human hepatoma cells (BEL-7402) were treated with basic FGF (b FGF) and wortmannin, and the activity of extracellular signal-regulated protein kinase B was detected by Western blot. Results Bcl-2 stimulated human hepatocellular carcinoma cells (BEL-7402) for 10 min at 25 ng / ml bFGF, and reached the highest PKC activity in the plasma membrane and cytoplasm. After Wortmannin pretreatment, the cell membrane and cytoplasmic PKB activity of human hepatocellular carcinoma cells (Bel-7402) were significantly decreased at all time points. Conclusion bFGF rapidly activates the PKB activity of human hepatocellular carcinoma cells (BEL-7402) through the phosphatidylinositol 3-kinase (PI3K) pathway.
