目的 :了解三七总皂苷 (PNS)及其单体 (Rg1、Rb1、Re+R1 )对造血祖细胞的增殖作用 ,以筛选出促进造血的有效单体及其浓度效应点。方法 :采用骨髓粒系、红系祖细胞 (CFU GM和 CFU E)半固体培养集落形成法 ,观察不同浓度的 PNS及其单体 (Rg1、Rb1、Re+R1 )对人造血祖细胞的刺激增殖作用。结果 :1 PNS对造血祖细胞有增殖作用 ,浓度为 8、4 0和 1 0 0 mg/ L时 ,CFU GM集落产率均明显高于对照组 (P均 <0 .0 1 ) ,提高率分别为 (32 .5± 6 .8) %、(5 9.3± 8.7) %和 (37.0± 1 .6 ) % ;CFU E集落增高在 PNS浓度为4 0 m g/ L时最为明显 ,提高率为 (33.3± 7.3) % (P<0 .0 1 )。 2 PNS单体对造血祖细胞有增殖作用 ,Rg1浓度为8和 4 0 mg/ L 时 ,CFU GM集落产率明显高于对照组 (P均 <0 .0 1 ) ,提高率分别为 (33.5± 3.8) %和 (5 9.7±4 .7) % ;Rb1浓度 4 0 m g/ L 时 ,CFU E集落产率明显高于对照组 (P<0 .0 1 ) ,提高率为 (4 7.3± 6 .3) % ;单体Re+R1对 CFU GM和 CFU E均无明显作用。结论 :1 PNS能够促进人骨髓粒系、红系造血祖细胞的增殖 ;2三七 Rg1和 Rb1是促进造血的有效单体
Objective : To understand the effect of Panax notoginseng saponins (PNS) and their monomers (Rg1, Rb1, Re+R1) on the proliferation of hematopoietic progenitor cells, so as to screen out the effective monomers and their concentration effect points for promoting hematopoiesis. METHODS: Semi-solid colony formation method using bone marrow granulocytic and erythroid progenitor cells (CFU GM and CFU E) was used to observe the stimulation of human hematopoietic progenitor cells with different concentrations of PNS and its monomers (Rg1, Rb1, Re+R1). Proliferation. RESULTS: 1 PNS had a proliferative effect on hematopoietic progenitor cells. The concentration of CFU GM was significantly higher than that of the control group (P < 0.01) when the concentrations were 8, 40, and 100 mg/L. They were (32. 5 ± 6.8)%, (5 9.3 ± 8.7)%, and (37.0 ± 1.6)%, respectively; the CFU E colonies increased most significantly when the PNS concentration was 40 mg/L. (33.3 ± 7.3) % (P < 0.01). 2 PNS monomer had a proliferative effect on hematopoietic progenitor cells. When Rg1 concentration was 8 and 40 mg/L, the CFU GM colony yield was significantly higher than that of the control group (P<0.01), and the increase rate was (33.5). ± 3.8)% and (5 9.7±4.7%)%; Rb1 concentration of 40 mg/L, the CFU E colony yield was significantly higher than that of the control group (P<0.01), and the increase rate was (47.3±). 6.3%; Monomer Re+R1 had no significant effect on CFU GM and CFU E. Conclusion : 1 PNS can promote the proliferation of human bone marrow granulocyte and erythroid hematopoietic progenitor cells; 2 R7 and Rb1 are effective monomers for promoting hematopoiesis
