目的:探讨护津方对细菌脂多糖(Lipopolysaccharide,LPS)诱导的急性肺损伤大鼠支气管肺泡灌洗液(Bronchoalveolar lavage fluid,BALF)中弹性蛋白酶(Neutrophil elastase,NE)及肺组织肿瘤坏死因子-αmRNA(Tumor necrosis factor-α mRNA,TNF-α mRNA)表达的影响。方法:40只雄性Wistar大鼠随机分为5组:正常对照组、LPS模型组、地塞米松组、护津方组,每组再分为4h和8h两个亚组。分别用地塞米松和护津方在LPS诱导ALI模型前预处理大鼠。分别采用酶联免疫吸附法和实时荧光定量PCR检测NE及TNF-α mRNA的表达。结果:两个时相点中,与模型组相比,护津方组BALF中NE含量均显著降低(P<0.01)。与模型组相比,8h组中,护津方组TNF-α mRNA表达均显著降低(P<0.01)。病理学观察,模型组大鼠肺组织出现大片出血及坏死,护津方组大鼠肺组织病理损伤程度较模型组减轻。结论:护津方能减轻内毒素致急性肺损伤大鼠肺组织损伤,对内毒素致急性肺损伤大鼠有保护作用,其机理可能与其能降低内毒素致急性肺损伤BALF中NE及肺组织TNF-α mRNA的表达有关。
Objective: To investigate the effect of Hujinfang on the expression of Neutrophil elastase (NE) and tumor necrosis factor - α (TNF - α) in bronchoalveolar lavage fluid (BALF) induced by Lipopolysaccharide (LPS) α mRNA (Tumor necrosis factor-α mRNA, TNF-α mRNA) expression. Methods: Forty male Wistar rats were randomly divided into 5 groups: normal control group, LPS model group, dexamethasone group and Xuanjinfang group. Each group was subdivided into 4 subgroups and 8h subgroups. Rats were pretreated with dexamethasone and Xuanjin before LPS-induced ALI. Enzyme-linked immunosorbent assay and real-time fluorescence quantitative PCR were used to detect the mRNA expression of NE and TNF-α. Results: Compared with the model group, the content of NE in BALF of the two groups was significantly decreased at both time points (P <0.01). Compared with the model group, the expression of TNF-α mRNA in the 8-h group was significantly decreased (P <0.01). Pathological observation, the model group rats lung tissue showed large hemorrhage and necrosis, the protective group of lung tissue pathological damage in rats than the model group. Conclusion: Hujin decoction can reduce lung injury induced by endotoxin-induced acute lung injury in rats, and has a protective effect on endotoxin-induced acute lung injury in rats. The mechanism may be related to its ability to reduce endotoxin induced acute lung injury in BALF NE and lung tissue TNF-α mRNA expression.
