[目的]探讨在CV-1细胞中肝细胞核因子3(HNF3)对Ⅱ型葡萄糖载体(GLUT2)表达的影响.[方法]利用DNA克隆技术将HNF3cDNA序列重组到表达载体pSV-sport上,GLUT2cDNA克隆到载体pGL3b上.通过CV-1细胞株的体外培养将HNF3转染CV-1培养细胞,应用荧光素酶活性测定和Western blot方法观察HNF3对GLUT2的启动子活性及蛋白质表达的影响.[结果]HNF3可增高GLUT2的荧光素酶活性及蛋白质表达水平.[结论]HNF3可能通过调节GLUT2的表达参与胰岛素对血糖的调节过程.
[Objective] To investigate the effect of HNF3 on the expression of GLUT2 in CV-1 cells. [Methods] The HNF3 cDNA sequence was recombined into the expression vector pSV-sport using DNA cloning technique. The GLUT2 cDNA clone HNF3 was transfected into CV-1 cells by in vitro culture of CV-1 cells, and the effect of HNF3 on the promoter activity and protein expression of GLUT2 was observed by luciferase activity assay and Western blot method. [Results] ] HNF3 can increase the GLUT2 luciferase activity and protein expression level. [Conclusion] HNF3 may be involved in the regulation of insulin on blood glucose by regulating the expression of GLUT2.
