增液润燥汤对干燥综合征NOD小鼠颌下腺中T、B淋巴细胞浸润及AQP5表达水平的影响

目的:观察增液润燥汤对干燥综合征(SS)NOD小鼠颌下腺中T、B淋巴细胞浸润情况以及AQP5表达水平的影响。方法:将40只8周龄NOD小鼠随机分为5组,即模型组,增液润燥汤低、中、高剂量组与羟氯喹组,同时选取8只同龄Balb/c小鼠作为正常组。增液润燥汤低、中、高剂量组分别给予10.5、21.0、42.0g/kg增液润燥汤灌胃;羟氯喹组给予60mg/kg羟氯喹灌胃;正常组与模型组给予同等剂量的去离子水灌胃。8周后,测量体质量后于麻醉下处死,摘取颌下腺、脾腺与胸腺称重并计算脏器指数;使用免疫组化法检测小鼠颌下腺中T、B淋巴细胞浸润量以及水通道蛋白5(AQP5)表达水平。结果:模型组小鼠颌下腺指数与正常组比较明显下降(P<0.05),药物干预后小鼠颌下腺指数变化不明显。模型组小鼠脾脏和胸腺指数与正常比较变化不明显。免疫组化结果显示,正常组小鼠颌下腺中未见有T与B淋巴细胞的浸润,模型组小鼠颌下腺中可见有大量的T、B淋巴细胞浸润;干预8周后,增液润燥汤各剂量组小鼠颌下腺中T、B淋巴细胞浸润量与模型组比较皆明显下降(P<0.05)。模型组小鼠AQP5的表达水平与正常组小鼠比较明显降低(P<0.05);干预8周后,增液润燥汤各剂量组小鼠颌下腺中AQP5的表达水平皆明显上升(P<0.05)。结论:NOD小鼠颌下腺中T和B淋巴细胞的浸润量明显增多、AQP5的表达明显减少;增液润燥汤能够有效抑制NOD小鼠颌下腺中T和B淋巴细胞浸润、促进AQP5的表达。 Objective: To observe the effect of Zeng Mo Run Decoction on T, B lymphocyte infiltration and AQP5 expression in submandibular gland of mice with Sjogren’s syndrome (SS). Methods: Forty NOD mice of 8 weeks old were randomly divided into 5 groups: model group, low, medium and high dose of ZYRT, and 8 groups of same age Balb / c mice as normal group. The rats in low, medium and high dosage group of adding liquid to moistening soup were given gavage of 10.5, 21.0 and 4.02.0g / kg, respectively. The hydroxychloroquine group was given 60mg / kg of hydroxychloroquine for gavage. The normal group and model group were given the same dosage Of deionized water gavage. After 8 weeks, the body weight was measured and sacrificed after anesthesia. The submandibular gland, spleen gland and thymus were removed and weighed and the organ index was calculated. The T, B lymphocyte infiltration in submandibular gland of mice and the expression of aquaporin 5 (AQP5) expression levels. Results: The index of submandibular gland in model group was significantly lower than that in normal group (P <0.05), and the index of submandibular gland in mice was not changed obviously after the intervention. The model group mice spleen and thymus index compared with the normal change was not obvious. Immunohistochemical results showed that there was no infiltration of T and B lymphocytes in the submandibular gland of mice in normal group. The infiltration of T and B lymphocytes in the submandibular gland of model mice was observed. After 8 weeks of intervention, The T and B lymphocyte infiltration in the submandibular gland of mice in each dose group were significantly decreased compared with the model group (P <0.05). The expression of AQP5 in the model group was significantly lower than that in the normal group (P <0.05). After 8 weeks of intervention, the expression of AQP5 in the submandibular gland of each group increased significantly (P <0.05 ). CONCLUSION: The infiltration of T and B lymphocytes in NOD mice submandibular glands significantly increased, while the expression of AQP5 decreased significantly. ZMYR could effectively inhibit the infiltration of T and B lymphocytes in submandibular glands of NOD mice and promote the expression of AQP5.
