目的 :应用微阵列技术探讨CDC10在星形细胞起源胶质瘤中的表达。方法 :术中收集 17例不同级的新鲜星形细胞起源胶质瘤标本 ,提取总RNA后逆转录成3 3 P标记的cDNA探针 ,与AtlascDNA微阵列杂交 ,获得相应基因表达谱 ,与正常脑组织的基因表达谱比较 ,并应用RT PCR验证结果。结果 :发现CDC10在所有肿瘤基因表达谱中均明显下调 ;经RT PCR验证 ,相对于正常脑组织的高mRNA丰度 ,所有肿瘤中该基因均下调甚至无表达。结论 :应用微阵列平行分析有助于发现已知基因的新功能。CDC10在星形细胞起源胶质瘤中表达下调 ,其功能有待进一步分析
Objective: To investigate the expression of CDC10 in astrocytic glioma using microarray technique. Methods: Seventeen fresh astrocytic origin glioma samples were collected during the operation. The total RNA was extracted and reversely transcribed into 3 3 P labeled cDNA probes. The cDNA probes were hybridized with Atlas cDNA microarrays to obtain the corresponding gene expression profiles. Brain tissue gene expression profiles were compared and RT PCR validation results were obtained. Results: CDC10 was found to be significantly down-regulated in all tumor gene expression profiles. All the tumors were downregulated or even underexpressed by RT-PCR as compared to normal brain tissue with high mRNA abundance. Conclusions: Parallel analysis using microarrays can help find novel functions of known genes. CDC10 is down-regulated in astrocytoma and its function needs further analysis
