目的:探讨分泌型磷脂酶A2(sPLA2)和过氧化物还原酶6(Prx6)蛋白在食管上皮癌变以及胎儿食管上皮发育过程中的表达及意义。方法:采用免疫组织化学ABC法检测河南食管癌高发区83例食管鳞状细胞癌(SCC)组织及癌旁正常组织、基底细胞过度增生、不典型增生和原位癌及65例3~9个月不同胎龄胎儿食管上皮发育分化过程中Prx6、sPLA2蛋白的表达。结果:成人正常食管上皮及各级病变组织中Prx6和sPLA2蛋白均有不同程度的表达,随病变程度加重,Prx6蛋白阳性表达率下降,而sPLA2蛋白阳性表达率升高(P<0.05);胎儿食管上皮分化发育过程中(3~9个月)Prx6蛋白阳性表达率与胎龄无相关性(P>0.05),sPLA2阳性表达率一直处于较高水平(P>0.05)。高分化食管癌组织Prx6和sPLA2蛋白阳性表达率均高于低分化食管癌组织(P<0.05)。结论:Prx6在食管上皮细胞分化诱导过程中起重要作用,sPLA2蛋白可能与食管上皮细胞的增殖有关。
Objective: To investigate the expression and significance of secreted phospholipase A2 (sPLA2) and peroxiredoxin 6 (Prx6) proteins in the carcinogenesis of esophageal epithelium and the development of fetal esophageal epithelium. Methods: 83 cases of esophageal squamous cell carcinoma (SCC) tissues, adjacent normal tissues, basal cell hyperplasia, dysplasia and carcinoma in situ and 65 cases of 3-9 Expression of Prx6 and sPLA2 proteins during the development and differentiation of fetal esophageal epithelium in different gestation months. Results: The expression of Prx6 and sPLA2 in normal esophageal epithelium and all kinds of diseased tissues were different in some extent. With the increasing degree of lesion, the positive rate of Prx6 protein decreased and the expression of sPLA2 protein increased (P <0.05) There was no correlation between Prx6 protein expression and gestational age (P> 0.05) during esophageal epithelial differentiation (3-9 months). The positive expression rate of sPLA2 was consistently high (P> 0.05). The positive rates of Prx6 and sPLA2 in well-differentiated esophageal cancer tissues were higher than those in poorly differentiated esophageal cancer tissues (P <0.05). Conclusion: Prx6 plays an important role in the induction of esophageal epithelial cells differentiation. The sPLA2 protein may be involved in the proliferation of esophageal epithelial cells.
