目的:提取五味子多糖,测定其含量并分析多糖的抗突变性,为研究五味子多糖提供实验参考。方法:水提醇沉法提取五味子多糖,0.1%活性碳脱色,苯酚-硫酸法测定多糖含量,进行小鼠姐妹染色单体交换及染色体畸变实验。结果:五味子多糖含量为85.56%,五味子多糖对环磷酰胺(CP)诱导的体细胞染色体畸变率及姐妹染色单体交换率具有抑制作用(P<0.05)。结论:苯酚-硫酸法简便、灵敏,可用于五味子多糖的测定;五味子多糖具有抗突变性。
Objective: To extract Schisandra chinensis polysaccharide, determine its content and analyze the anti-mutagenicity of polysaccharide, and provide experimental reference for the research of Schisandra chinensis polysaccharide. Methods: Water extraction and alcohol precipitation were used to extract Schisandra chinensis polysaccharides, 0.1% activated carbon was decolorized, phenol-sulfuric acid method was used to determine polysaccharide content, and sister sister chromatid exchange and chromosomal aberration experiments were performed. Results: The content of polysaccharides in Schisandra chinensis was 85.56%. Schisandra chinensis polysaccharides inhibited the chromosome aberration rate and the sister chromatid exchange rate induced by cyclophosphamide (CP) (P<0.05). Conclusion: The phenol-sulfuric acid method is simple and sensitive, and can be used for the determination of polysaccharides of Schisandra chinensis. Schisandra chinensis polysaccharide has anti-mutagenicity.
