以云南芥菜型油菜部分品种及雄性不育系S187A、恢复系S187C为材料,进行花药培养诱导胚状体试验。结果表明,培养效率受材料基因型的影响较大;采用液体漂浮分步培养法较固体培养法诱导频率高,前者的胚状体诱导频率达42.1%,而采用固体培养方法仅为3.2%。在液体漂浮分步培养程序中加入适当浓度的多效唑,有利于提高胚状体的诱导频率,其中有的处理胚状体诱导频率达到214%。
Some embryos of Brassica juncea and male sterile line S187A and restorer line S187C were used as materials to induce embryoid body in anther culture. The results showed that the culture efficiency was greatly influenced by the genotype of the material. The induction rate of the former embryoid body was 42.1%, which was higher than that of the solid culture method by liquid floating fractional culture method, while the solid culture method was only 3 .2%. Adding appropriate concentrations of paclobutrazol in the step-by-step liquid-flooding process is beneficial to improve the induction frequency of embryoid bodies, and some of them deal with embryoid body induction frequency of 214%.
