目的 研究成釉细胞瘤 (ameloblastoma ,AB)和牙源性角化囊肿 (odontogenickeratocyst,OKC)中hTERTmRNA的表达 ,探讨其在AB、OKC中的发生、发展及其生物学意义。方法 采用mRNA原位杂交法对口腔颌骨内 5 4例AB、16例OKC及 7例口腔正常粘膜 ,进行了hTERTmRNA的检测。结果 94 4 % (5 1/5 4 )AB、87 5 % (14 /16 )OKC及 1/7正常口腔粘膜有hTERTmRNA表达 ,3组相比差异有显著性 (P <0 0 0 1)。hTERT与AB不同组织学类型、病损发生部位、单房与多房无关 (P >0 0 5 ) ,在AB外周细胞及星网状多边形细胞中有表达 ,在角化退变样细胞及颗粒样细胞中缺乏表达。结论 ①hTERT活化在AB及OKC的发生、发展中起重要作用 ;②hTERT活性阳性率与病变中的细胞分化 ,临床生物学行为有关 ,随着组织学分级增高hTERTmRNA阳性率及信号强度均增高。目的 研究成釉细胞瘤 (ameloblastoma ,AB)和牙源性角化囊肿 (odontogenickeratocyst,OKC)中hTERTmRNA的表达 ,探讨其在AB、OKC中的发生、发展及其生物学意义。方法 采用mRNA原位杂交法对口腔颌骨内 5 4例AB、16例OKC及 7例口腔正常粘膜 ,进行了hTERTmRNA的检测。结果 94 4 % (5 1/5 4 )AB、87 5 % (14 /16 )OKC及 1/7正常口腔粘膜有hTERTmRNA表达 ,3组相比差异有显著性 (P <0
Objective To study the expression of hTERT mRNA in ameloblastoma (AB) and odontogenic keratocyst (OKC), and to investigate its occurrence, development and biological significance in AB and OKC. Methods The mRNA of hTERTmRNA was detected by in situ hybridization in 54 oral mucosa, 16 cases of OKC and 7 cases of oral normal mucosa. Results There was significant difference in hTERT mRNA expression between 94 4% (5 1/5 4) AB, 87 5% (14/16) OKC and 1/7 normal oral mucosa. There was significant difference between the three groups (P <0.01). hTERT and AB different histological types, the lesion site, single room and multiple rooms has nothing to do (P> 0.05), in the AB peripheral and reticular polygonal cells in keratinocyte degeneration and granulocyte-like Lack of expression in cells. Conclusion ① The activation of hTERT plays an important role in the development of AB and OKC. ② The positive rate of hTERT activity is related to the cell differentiation and clinical biological behavior. The positive rate and signal intensity of hTERT mRNA are increased with the increase of histological grade.
