[目的]观察红三叶草异黄酮对去卵巢大鼠骨密度及骨代谢的影响。[方法]将55只体重(250±10)g雌性SD大鼠随机分为5组:假手术组(灌胃蒸馏水)、模型组(灌胃蒸馏水)、雌激素组(乙烯雌酚)、红三叶草异黄酮处理组(高、低剂量)。除假手术组外,其他各组均切除双侧卵巢,假手术组行相同手术操作,但不切除卵巢。各组动物均在相同的环境中饲养,自由摄食和饮水,造模1周后开始喂药。实验8周后对大鼠双股骨骨密度(BMD)和骨矿含量(BMC)进行检测,心脏取血测定血清抗酒石酸酸性磷酸酶(TRACP)、碱性磷酸酶(AKP)和骨钙素(BGP)含量。[结果]与模型组比较,红三叶草异黄酮高剂量组能够增加去卵巢大鼠BMD和BMC,降低血清抗酒石酸酸性磷酸酶、碱性磷酸酶活性和骨钙素含量。[结论]红三叶草异黄酮不仅可以维持去卵巢大鼠骨密度,而且可以替代雌激素对骨代谢有一定的调节作用。
[Objective] To observe the effects of red clover isoflavone on bone mineral density and bone metabolism in ovariectomized rats. [Methods] Fifty-five female Sprague-Dawley rats weighing 250 ± 10 g were randomly divided into 5 groups: sham operation group (fed with distilled water), model group (fed with distilled water), estrogen group Clover isoflavones treatment group (high and low dose). Except for the sham operation group, all the other groups were resected with bilateral ovaries. The sham operation group performed the same operation but did not remove the ovary. Animals in each group were kept in the same environment, fed freely and drinking water, and started feeding after 1 week. Eight weeks after the experiment, bone mineral density (BMD) and bone mineral content (BMC) of both femurs were measured. Blood samples were collected for determination of serum TRACP, AKP and osteocalcin BGP) content. [Result] Compared with the model group, the red clover isoflavone high dose group could increase the BMD and BMC of ovariectomized rats and decrease the serum tartrate-resistant acid phosphatase, alkaline phosphatase activity and osteocalcin levels. [Conclusion] Red clover isoflavone not only can maintain the bone density of ovariectomized rats, but also can replace estrogen to regulate bone metabolism.
