应用电子吸收光谱、溴化乙锭(EB)荧光猝灭光谱、粘度测定及琼脂糖凝胶电泳等方法研究了配合物[Cu(IP)(L-Leu)(H2O)]ClO4(1)和[Cu(IP)(L-Trp)(H2O)]ClO4.1.5H2O(2)(IP=咪唑并[5,6-f][1,10]邻菲咯啉,L-Leu=L-亮氨酸,L-Trp=L-酪氨酸)与DNA的相互作用。分析结果表明,配合物通过插入模式与DNA作用,在还原剂维生素C存在的条件下通过羟基自由基机理对DNA进行切割,作用大小次序为:配合物1>2。
The effects of complex [Cu (IP) (L-Leu) (H2O)] ClO4 (1) and (Cu2 +) on the fluorescence spectra of the complexes were studied by electron absorption spectroscopy, fluorescence spectrophotometry of ethidium bromide (EB), viscosity measurement and agarose gel electrophoresis. [L-Trp (H2O)] ClO4.1.5H2O (2) (IP = imidazo [5,6-f] [1,10] phenanthroline, L-Leu = L- Amino acid, L-Trp = L-tyrosine) and DNA. The results showed that the DNA was cleaved by the hydroxyl radical mechanism in the presence of reducing agent vitamin C through the interaction between insertion mode and DNA. The order of action was: complex 1> 2.
