目的:观察五味子乙素(Sch-B)对二氧化硅致大鼠矽肺中基质金属蛋白酶2(matrix metallopro-teinase,MMP-2)和其抑制物基质金属蛋白酶组织抑制因子1(tissue inhibitor of matrix metalloproteinase,TIMP-1)含量的影响。方法:96只大鼠随机分为对照组、染尘组和Sch-B干预组,每组32只,采用暴露式气管内注入SiO2混悬液法建立大鼠矽肺模型,造模后1 d灌胃给予Sch-B干预治疗,分别在药物干预3,7,14,28 d时处死大鼠,取其肺组织,HE染色检测肺组织病理改变;ELISA法测定大鼠肺组织中MMP-2和TIMP-1蛋白的含量。结果:与对照组相比,染尘组MMP-2和TIMP-1蛋白含量在3,7,14,28 d时明显增高(P<0.01);与染尘组相比,Sch-B干预组MMP-2和TIMP-1蛋白含量均明显降低(P<0.01),但仍显著高于对照组(P<0.01),差异有统计学意义。结论:Sch-B对二氧化硅致大鼠矽肺中不同时间点肺组织MMP-2和TIMP-1的表达均有一定程度的抑制作用,能在一定程度上调整MMP-2和TIMP-1的比值使其趋于平衡,具有减轻二氧化硅致矽肺大鼠肺纤维化的作用。
OBJECTIVE: To observe the effects of Schisandrin B on the expression of matrix metallopro-tease (MMP-2) and inhibitor of matrix metalloproteinase-1 metalloproteinase, TIMP-1) content. Methods: Ninety-six rats were randomly divided into control group, Dian Dian group and Sch-B intervention group, with 32 rats in each group. Silicosis model was established by exposure to endobronchial SiO2 suspension. The rats were sacrificed at 3, 7, 14, and 28 days after drug intervention, respectively. The lung tissues of the rats were sacrificed and the pathological changes of the lungs were detected by HE staining. The expressions of MMP-2 and TIMP-1 protein content. Results: Compared with the control group, the protein levels of MMP-2 and TIMP-1 in the dust-exposed group were significantly increased at 3, 7, 14 and 28 days (P <0.01) The protein levels of MMP-2 and TIMP-1 were significantly decreased (P <0.01), but still significantly higher than those of the control group (P <0.01). The difference was statistically significant. CONCLUSION: Sch-B can inhibit the expression of MMP-2 and TIMP-1 in lung tissue of silicosis rats at different time points to a certain extent, and can regulate MMP-2 and TIMP-1 to a certain extent Ratio to make it more balanced, with silica to reduce silica-induced pulmonary fibrosis in rats.
