观测SH1对ADP,AA,Colagen诱导的兔血小板聚集及TXB2和6-keTO-PGFIα生成的影响。用比浊法测定了透骨草提取的单体生物碱结晶SH1体外对兔血小板聚集的影响,用放免法测定TXB2和6-keTO-PGFIα的含量。结果:SH10.8~3.0mmol/L范围内明显抑制AA,ADP,Colagen诱导的兔血小板聚集。SH1对三种诱导剂的最大抑制率分别为62.16%,45.25%,53.67%。大剂量组明显加快ADP诱导的血小板聚集后的解聚速度,SH1显著延长Colagen的诱导起聚时间。其抑制作用有明显的量效关系,对AA诱导的TXB2产生明显抑制作用,可使6-keTO-PGFIα的含量略有增加,但6-keTO-PGFIα/TXB2比值显著增大
To observe the effect of SH1 on platelet aggregation induced by ADP, AA and Colagen and the generation of TXB2 and 6-keTO-PGFIα. The turbidimetric method was used to determine the effect of the extracellular alkaloid extracted from Septumria divaricata on platelet aggregation in vitro. The content of TXB2 and 6-keTO-PGFIα were determined by radioimmunoassay. Results: Rabbit platelet aggregation induced by AA, ADP and Colagen was significantly inhibited in SH10.8 ~ 3.0mmol / L. The maximum inhibitory rates of SH1 to the three inducing agents were 62.16%, 45.25% and 53.67%, respectively. The high-dose group significantly accelerated the rate of ADP-induced platelet aggregation, and SH1 significantly prolonged Colagen-induced polymerization. The inhibitory effect was significantly dose-effect relationship, which could inhibit the TXB2 induced by AA and increase the content of 6-keTO-PGFIα slightly, but the ratio of 6-keTO-PGFIα / TXB2 was significantly increased
