目的 研究gp130单克隆抗体B S12的生物学特性。方法 用细胞计数和3 H TdR掺入的方法观察人多发性骨髓瘤细胞株XG 1和XG 2在抗gp130的单抗B S12与它们的Fab片段和F(ab′) 2 片段作用下 ,细胞增殖和生长情况 ;此外 ,也观察gp130单抗B P8以及IL 6和IL 6R单抗B E8和B R6对B S12单抗在这 2个细胞株上作用的影响。结果 B S12可刺激XG 1和XG 2细胞增殖 ,并维持它们的长期生长 ,但B S12的这些作用有赖于它保持完整的二价结合力。B E8和B R6对B S12的作用无影响 ,B P8则可协同增强B S12的作用。结论 B S12为刺激型抗gp130的单抗 ,可起着类似于IL 6的作用。
Objective To study the biological characteristics of gp130 monoclonal antibody B S12. Methods Human multiple myeloma cell lines XG 1 and XG 2 were observed by cell counting and incorporation of 3 H TdR. Cells were treated with anti-gp130 mAb B S12 and their Fab and F (ab ’) 2 fragments Proliferation and growth. In addition, the effects of gp130 mAb B P8 and IL6 and IL6R mAb B E8 and B R6 on the effect of B S12 mAb on these two cell lines were also observed. Results B S12 stimulates the proliferation of XG 1 and XG 2 cells and maintains their long-term growth, but these effects of B S12 depend on it maintaining intact bivalent binding. B E8 and B R6 have no effect on B S12, and B P8 synergistically enhances B S12. Conclusion B S12 is a monoclonal antibody against stimulated gp130, which may play a role similar to IL 6.
