目的探讨气营传变对兔舌背黏膜TLR4(Toll like receptors 4,Toll样受体4)和E-cad(E-cadherin,E-钙粘蛋白)表达的影响。方法利用LPS(Lipopolysaccharide,脂多糖)对兔耳缘静脉进行注射,建立气分证和营分证模型,以白虎汤和清营汤对证干预。HE染色及免疫组化研究TLR4和E-cad表达水平。结果与对照组相比,营分证组舌背黏膜显著增厚(P<0.01),TLR4在气分证与营分证组中的表达均显著升高(P<0.01),气分证及营分证组中舌背黏膜E-cad表达显著降低(P<0.01),清营汤可显著逆转营分证组舌背黏膜TLR4表达(P<0.05)和E-cad表达(P<0.01),而白虎汤效果不显著(P>0.05)。结论气营传变过程影响兔舌背黏膜厚度,与TLR4、E-cad表达变化有关。
Objective To investigate the effect of gas-qi-qing-qing-ling on expression of TLR4 (Toll like receptor 4) and E-cadherin (E-cadherin) in rabbit tongue mucosa. Methods Lipopolysaccharide (LPS) was used to inject the rabbits’ veins of the rabbits to establish the Qi-Differentiation and Camp-Diary Syndrome models. The expression of TLR4 and E-cad was examined by HE staining and immunohistochemistry. Results Compared with the control group, the tongue mucosa of Yingdiuji group was significantly thicker (P <0.01), and the expression of TLR4 was significantly higher in Qi and Zhengyingzheng groups (P <0.01) The expression of E-cadherin in tongue back mucosa of Yingjian Syndrome group was significantly decreased (P <0.01), Qingying Decoction significantly reversed the expression of TLR4 (P <0.05) and E-cadherin (P <0.01) , While the effect of Baihu Tang was not significant (P> 0.05). Conclusions Gas migration process affects the thickness of tongue mucosa in rabbits and is related to the changes of TLR4 and E-cad.
