目的:建立采用高效液相色谱同时测定维吾尔药阿纳其根中2个N-烷基类成分含量的方法,为制定阿纳其根质量标准提供依据。方法:采用LC-20A型高效液相色谱仪,Agilent TC-C_(18)色谱柱(4.6 mm×150 mm,5μm),以甲醇-水(70:30)为流动相,流速0.8 mL·min~(-1),检测波长243 nm,柱温25℃。结果:十-2E,4E-二烯酸-4-羟基苯乙胺和十二-2E,4E-二烯酸-4-羟基苯乙胺质量浓度分别在30.6~122.4 mg·L~(-1)和17.2~68.8 mg·L~(-1)范围内呈良好线性关系,平均加样回收率(n=6)分别为99.90%(RSD=1.60%)和100.4%(RSD=0.85%);3批阿纳其根药材中两者含量范围分别为0.046%~0.054%和0.035%~0.039%(n=3)。结论:经方法学验证,该方法可为阿纳其根药材质量控制提供一定的参考。
OBJECTIVE: To establish a method for the simultaneous determination of two N-alkyl components in Aniqi root of Uyghur medicine by high performance liquid chromatography (HPLC), and to provide the basis for establishing the quality standard of Anazaki root. Methods: The mobile phase consisted of methanol-water (70:30) with a flow rate of 0.8 mL · min using LC-20A high performance liquid chromatography with Agilent TC-C 18 column (4.6 mm × 150 mm, 5 μm) ~ (-1). The detection wavelength was 243 nm and the column temperature was 25 ℃. Results: The mass concentration of 10-E, 4E-dienoic acid and 4-hydroxyphenethylamine of 12-E, 4E-dienoic acid were 30.6-122.4 mg · L -1 (N = 6) were 99.90% (RSD = 1.60%) and 100.4% (RSD = 0.85%), respectively, in the range of 17.2-68.8 mg · L -1. The content of the two batches in three batches of Anatomical herbs were 0.046% ~ 0.054% and 0.035% ~ 0.039%, respectively (n = 3). Conclusion: This method can be used as a reference for the quality control of Anatain roots by methodological verification.
