目的:研究中药虎杖中蒽醌的荧光性质及其与自由基清除活性的相关性。方法:采用70%乙醇提取、乙酸乙酯萃取、碱水解和酸沉淀,自虎杖中分离虎杖蒽醌,采用荧光扫描确定其激发波长和发射波长,考察pH、温度及光照时间对其荧光强度的影响。采用DPPH法测定其对自由基清除活性。结果:虎杖蒽醌的最小激发波长和最大发射波长分别为369nm和575nm。pH碱性、提高温度和光照均可降低其荧光强度。虎杖蒽醌的自由基清除率与其荧光强度在0~800μg/m l浓度范围内呈线性相关(r=0.9849,P<0.01)。结论:虎杖蒽醌具有光活化作用,在中性、酸性、低温和避光条件下稳定,可通过荧光测定来判断其自由基清除活性。
Objective: To study the fluorescence properties of anthraquinones from Polygonum cuspidatum and its correlation with free radical scavenging activity. Methods: Polygonatum anthrolin was separated from Polygonum cuspidatum by 70% ethanol extraction, ethyl acetate extraction, alkali hydrolysis and acid precipitation. Fluorescence scanning was used to determine the excitation and emission wavelengths. The effects of pH, temperature and illumination time on the fluorescence intensity influences. DPPH method was used to determine the free radical scavenging activity. Results: The minimum excitation wavelength and maximum emission wavelength of Polygonatum anthraquinone were 369nm and 575nm, respectively. pH alkaline, increase the temperature and light can reduce its fluorescence intensity. The free radical scavenging rate of anthraquinones in Polygonum cuspidatum was linearly correlated with its fluorescence intensity in the range of 0 ~ 800μg / ml (r = 0.9849, P <0.01). Conclusion: Polygonatum anthraquinone has the function of light activation and is stable under neutral, acid, low temperature and dark conditions. Its free radical scavenging activity can be judged by fluorescence measurement.
