本文构建了一个包含 16 12 8个克隆的稻瘟菌基因组的TAC文库 ,74%的克隆含有外源插入片段 ,其插入片段大小平均为 5 9kb ,该库相当于稻瘟菌基因组的 18倍。本文并对文库的代表性进行了评价 ,采用 3个单拷贝的标记作为探针筛库 ,分别得到 16 ,17和 16个阳性克隆 ,这与估算的该库的覆盖率是一致的 ;从MH18S1为探针所筛选的阳性克隆中 ,随机挑取 8个并对其限制性酶切图谱进行分析 ,结果表明 :其组成的重叠群跨度为 15 3kb ,未发现嵌合和缺失现象。以上结果表明 ,此TAC文库适合用于稻瘟菌基因组的分析。
In this paper, we constructed a TAC library containing 16128 clones of Magnaporthe grisea, and 74% of the clones contained exogenous inserts with an average insert size of 59kb, which is 18 times of the Magnaporthe grisea genome. In this paper, we also evaluated the representativeness of the library. Using three single-copy markers as probe screens, 16, 17 and 16 positive clones were obtained respectively, which is consistent with the estimated coverage of the library. From MH18S1 For the positive clones screened by the probes, 8 were randomly selected and their restriction maps were analyzed. The results showed that the span of the contigs was 15 3kb, and no chimerism and deletion were found. The above results show that this TAC library is suitable for the genome analysis of Magnaporthe grisea.
