在放免分析中,如何选择优良的分离剂,将抗原抗体复合物(B)及游离抗原(F)分开,是建立完善的放免方法的重要环节。理想的分离剂应具有下列条件;(1)B和F分离快速而完全;(2)操作简便,试剂来源容易,价格低,便于推广;(3)不受其它试剂的干扰,重复性良好。常用的分离剂有离子交换树脂、硅镁吸附剂、活性炭、第二抗体、硫酸胺、聚乙二醇(PEG)等,其中最常用者为PEG及大分子包被活性炭,PEG法
