目的建立测定安神颗粒中金丝桃苷和芦丁的高效液相色谱法。方法采用Dimark十八烷基键合硅胶色谱柱(200mm×4.6mm,5μm);柱温:40℃;流动相:甲醇-1%冰醋酸(28∶72);体积流量:1.0mL/min;检测波长:359nm;进样量:20μL。结果金丝桃苷和芦丁在2.246~5.242μg/mL和2.110~4.922μg/mL与峰面积呈良好的线性关系,r分别为0.9999和0.9998。平均回收率分别为101.4%、99.9%。结论用此法测定安神颗粒中金丝桃苷和芦丁简单易行,结果准确。
Objective To establish a HPLC method for the determination of hyperin and rutin in Anshen granules. Methods Dimark octadecyl-bonded silica gel column (200mm×4.6mm, 5μm); column temperature: 40°C; mobile phase: methanol-1% acetic acid (28:72); volumetric flow rate: 1.0mL/min; Detection wavelength: 359 nm; injection volume: 20 μL. Results There was a good linear relationship between hyperin and rutin in the range of 2.246 to 5.242 μg/mL and 2.110 to 4.922 μg/mL, with r values of 0.9999 and 0.9998, respectively. The average recoveries were 101.4% and 99.9%, respectively. Conclusion The method is simple and accurate for the determination of hyperin and rutin in Anshen granules.
