目的阐明WNK4基因G1816T多态性与原发性高血压的关系。方法2001年3~10月选择中国东北高血压病高发地区辽宁省彰武县采集的259例原发性高血压患者和235名健康对照组的血样应用聚合酶链反应-限制性长度片段长度多态性分析(PCR-RFLP)方法检测G1816T多态,结果经测序验证,同时检测所有人群的血糖、血脂等生化指标。结果原发性高血压组WNK4基因G1816T多态T等位基因频率显著高于对照组(25·9%对20·2%,P=0·035);AS和SS基因型与收缩压及舒张压升高密切相关(P分别为0·041和0·032)。结论WNK4基因G1816T多态与高血压密切相关。
Objective To elucidate the relationship between WNK4 gene G1816T polymorphism and essential hypertension. Methods From March to October 2001, blood samples from 259 essential hypertension patients and 235 healthy controls collected from Zhangwu County of Liaoning Province in northeastern China with high prevalence of hypertension were analyzed by polymerase chain reaction - restriction fragment length Polymorphism of G1816T was detected by PCR-RFLP. The results were verified by sequencing. At the same time, biochemical indexes such as blood glucose and blood lipids were detected in all populations. Results The frequency of allele G1816T polymorphism of WNK4 gene was significantly higher in patients with primary hypertension than in controls (25.9% vs 20.2%, P = 0.035). The genotypes of AS and SS were correlated with systolic and diastolic Pressure rise is closely related (P = 0.041 and 0.032, respectively). Conclusion WNK4 gene G1816T polymorphism is closely related to hypertension.
