目的探讨右美托咪定(Dex)混合氯胺酮对新生大鼠离体海马细胞凋亡的影响。方法 SD新生大鼠(出生7 d)海马神经元体外培养6 d,采用随机数字表法分为对照组(C组),100μmol/L氯胺酮组(K组),10μg/ml Dex组(D组),10μg/ml Dex混合100μmol/L氯胺酮组(DK组),各6只。孵育24 h后采用免疫组化法检测Caspase-3蛋白的表达,双染法检测神经元凋亡。结果与C组比较,K组的海马细胞凋亡率和Caspase-3表达水平升高(P<0.05);与C组比较,D组海马细胞凋亡率和Caspase-3表达水平差异无统计学意义(P>0.05);与K组比较,DK组和D组的海马细胞凋亡率和Caspase-3表达水平降低(P<0.05)。结论 Dex能减轻氯胺酮诱导的新生大鼠离体海马细胞凋亡。
Objective To investigate the effect of dexmedetomidine mixed ketamine on apoptosis of hippocampus in neonatal rats. Methods Hippocampal neurons of SD neonatal rats (born 7 days after birth) were cultured in vitro for 6 days and were divided into control group (C group), K (100 μmol / L) group, Dex group ), 10μg / ml Dex mixed 100μmol / L ketamine group (DK group), 6 mice in each group. After incubation for 24 h, the expression of Caspase-3 protein was detected by immunohistochemistry and apoptosis was detected by double staining. Results Compared with group C, the apoptotic rate of hippocampus and the expression of Caspase-3 in group K were significantly increased (P <0.05). Compared with group C, there was no significant difference in apoptotic rate and Caspase-3 expression in group D (P> 0.05). Compared with K group, the apoptotic rates of hippocampus and the expression of Caspase-3 in DK group and D group decreased (P <0.05). Conclusion Dex can reduce ketamine-induced apoptosis in isolated hippocampal cells of neonatal rats.
