目的:观察环孢素A(Cyclosporin A,CsA)对大鼠磨牙牙龈结缔组织面积,血管数量,血管面积以及层粘连蛋白(laminin,LN)表达的影响。方法:建立CsA大鼠牙龈增生模型。制备下颌第一磨牙石蜡切片,分别进行HE染色,LN免疫组织化学染色。定量分析牙龈结缔组织面积、血管面积、5个视野血管数量计数。观察牙龈结缔组织内血管基膜LN的表达。结果:①组织测量结果:实验组颊侧牙龈结缔组织面积大于对照组,实验组颊舌侧牙龈结缔组织血管数量总数和均数多于对照组,实验组颊舌侧牙龈结缔组织中的血管总面积占牙龈结缔组织总面积的百分比大于对照组(各组数据P﹤0.05)②免疫组化结果:LN阳性表达于血管基膜、上皮基膜等部位。对血管基膜上LN阳性的血管进行分区域计数,实验组的总数和均数均大于对照组(P﹤0.05),且实验组血管基膜上LN表达强度上调。结论:①.CsA使大鼠牙龈结缔组织内血管化作用增强。②CsA使大鼠牙龈结缔组织内血管基底膜上LN表达增多。
Objective: To observe the effects of cyclosporin A (CsA) on gingival connective tissue area, blood vessel volume, vascular area and laminin (LN) expression in rat molar. Methods: A rat model of gingival hyperplasia was established. Paraffin sections of mandibular first molar were prepared and stained with HE and LN respectively. Quantitative analysis of gingival connective tissue area, vascular area, five visual field vascular count. The expression of vascular basement membrane LN in gingival connective tissue was observed. Results: ① The results of tissue measurement: The area of gingival connective tissue in the experimental group was larger than that in the control group. The total number and average number of gingival connective tissue blood vessels in the experimental group were more than those in the control group. The vascular total gingival connective tissue The area of gingival connective tissue area accounted for more than the control group (P <0.05 for each group) ② immunohistochemistry results: LN positive expression in vascular basement membrane, epithelial basement membrane and other parts. The number of LN-positive vessels in the basal lamina was counted in the sub-region. The total number and mean number of the experimental groups were all higher than those of the control group (P <0.05). The LN expression intensity of the basilar membrane in the experimental group was increased. Conclusions: ①CsA enhances vascularization in connective tissue of rats. ②CsA increased the expression of LN in the vascular basement membrane in rat gingival connective tissue.
