[目的]探讨沙立度胺(thalidomide,TD)及其对映体(S-thalidomide)S-TD对人非小细胞肺癌系NCI-H460细胞增殖的影响。[方法]体外培养NCI-H460细胞,采用MTT比色法测定不同质量浓度的TD、S-TD对NCI-H460细胞增殖的影响;采用流式细胞仪检测对细胞周期的影响;DAPI染色观察诱导细胞凋亡作用。[结果]在10.0~200.0μg/ml范围内,TD、S-TD作用于NCI-H460细胞24h可抑制细胞增殖、48h显著抑制细胞增殖,而在高浓度时(200.0μg/ml)S-TD的抑制作用弱于TD;TD、S-TD作用NCI-H460细胞24h、48h、72h,TD的抑制率分别为25.69%、41.92%、7.57%,S-TD为14.77%、33.24%、8.46%。200.0μg/mlTD和S-TD处理NCI-H460细胞48h后,G2/M期细胞比例显著增高,S期细胞比例下降。DAPI染色结果发现细胞数量明显减少,但未出现核固缩、核边集等凋亡形态学改变。[结论]TD和S-TD在一定浓度范围对NCI-H460细胞具有抗增殖作用,但S-TD的抗增殖活性稍弱于TD。抑制增殖作用可能与阻滞G2/M期细胞周期有关,DAPI染色结果未显示诱导细胞凋亡作用。
[Objective] To investigate the effects of thalidomide (TD) and its S-thalidomide S-TD on the proliferation of human non-small cell lung cancer cell line NCI-H460. [Methods] NCI-H460 cells were cultured in vitro. The effects of different concentrations of TD and S-TD on the proliferation of NCI-H460 cells were determined by MTT colorimetric assay. The cell cycle was detected by flow cytometry. DAPI staining Apoptosis. [Result] The proliferation of NCI-H460 cells was inhibited by TD and S-TD in the concentration range of 10.0 ~ 200.0μg / ml for 24 hours, and significantly inhibited by 48 hours. However, at high concentration (200.0μg / ml) The inhibitory rates of TD and S-TD on NCI-H460 cells were 25.69%, 41.92% and 7.57% at 24 h, 48 h and 72 h after TD and 14.77%, 33.24% and 8.46% . After treated with 200.0μg / ml of TD and S-TD for 48h, the proportion of cells in G2 / M phase increased significantly and the proportion of cells in S phase decreased. DAPI staining results showed that the number of cells was significantly reduced, but no nuclear pyknosis and nuclear bundles were observed. [Conclusion] TD and S-TD have antiproliferative effect on NCI-H460 cells in a certain concentration range, but the antiproliferative activity of S-TD is slightly weaker than that of TD. Inhibition of proliferation may be related to blocking the G2 / M cell cycle, DAPI staining results did not show the induction of apoptosis.
