目的探讨肿瘤相关巨噬细胞(TAMs)、血管内皮生长因子(VEGF)在高级别人膀胱癌组织中的表达特征。方法选取2014年4月至2016年4月行全膀胱切除术的30例高级别膀胱癌患者作为研究对象。取切除标本的膀胱肿瘤组织和膀胱正常组织,通过免疫组化方法用CD68标记巨噬细胞(CD68+),用CD34标记微血管密度(MVD),进行VEGF表达、TAMs浸润程度、MVD计数的检测,对比、分析检测结果。结果与正常膀胱黏膜相比,膀胱癌组织的CD68+TAMs表达等级明显上调(P<0.01)。膀胱癌组织的VEGF阳性表达率、MVD计数均显著高于正常膀胱黏膜(P均<0.01)。Pearson相关性检验显示,膀胱癌组织中VEGF表达、MVD计数分别与CD68+TAMs表达均呈显著正相关关系(r=0.896,P<0.01;r=0.908,P<0.01)。结论 VEGF和TAMs在高级别人膀胱癌组织中均呈高表达,VEGF与TAMs呈显著正相关,进行VEGF和TAMs检测能够为临床判定膀胱癌进展情况、预测患者预后提供可靠依据。
Objective To investigate the expression of tumor-associated macrophages (TAMs) and vascular endothelial growth factor (VEGF) in high-grade human bladder cancer. Methods Thirty patients with high-grade bladder cancer undergoing total cystectomy from April 2014 to April 2016 were selected as the study subjects. Tissue samples of bladder tumor and normal bladder tissues were obtained. CD68-labeled macrophages (CD68 +) and CD34-labeled microvessel density (MVD) were used to detect the expression of VEGF, the infiltration of TAMs and the count of MVD. , Analyze the test results. Results Compared with normal bladder mucosa, the expression of CD68 + TAMs in bladder cancer tissues was significantly increased (P <0.01). The positive expression rates of VEGF and MVD in bladder cancer tissues were significantly higher than those in normal bladder mucosa (all P <0.01). Pearson correlation test showed that the expression of VEGF and MVD in bladder cancer were positively correlated with the expression of CD68 + TAMs (r = 0.896, P <0.01; r = 0.908, P <0.01). Conclusions Both VEGF and TAMs are highly expressed in high-grade bladder cancer tissues. VEGF and TAMs have a significant positive correlation. The detection of VEGF and TAMs can provide a reliable basis for judging the progress of bladder cancer and predicting the prognosis of patients.
