目的:通过观察骨康含药血清对成骨细胞OPG、RANKL表达的影响,探讨其治疗骨质疏松的作用机制。方法:体外分离、培养成骨细胞,实验分为3组:正常血清组、中药骨康血清组和雌二醇含药血清组。采用酶联免疫吸附法,检测成骨细胞OPG和RANKL的表达。结果:中药骨康含药血清组RANKL含量显著低于空白对照组(P<0.05),而OPG分泌量显著高于空白对照组(P<0.05);中药骨康含药血清组与雌二醇含药血清组OPG和RANKL的含量无明显差异。结论:在去势状态下,中药骨康可能是抑制RANKL分泌,上调OPG分泌,而达到治疗骨质疏松症的作用。
OBJECTIVE: To observe the effects of GuKang drug-containing serum on the expression of OPG and RANKL in osteoblasts and explore the mechanism of osteoporosis treatment. METHODS: Osteoblasts were isolated and cultured in vitro. The experiment was divided into 3 groups: normal serum group, traditional Chinese medicine Gukang serum group and estradiol drug-containing serum group. The expression of OPG and RANKL in osteoblasts was detected by enzyme-linked immunosorbent assay. Results: The content of RANKL in the serum of Gukang medicine-containing group was significantly lower than that of the blank control group (P<0.05), while the secretion of OPG was significantly higher than that of the blank control group (P<0.05); the traditional Chinese medicine Gukang medicine-containing serum group and estradiol There was no significant difference in the contents of OPG and RANKL in the drug-containing serum group. Conclusion: In the state of castration, Gukang may suppress the secretion of RANKL and up-regulate the secretion of OPG to achieve the treatment of osteoporosis.
