以小鼠脾淋巴细胞转化,白介素2(IL-2)产生,胞内游离钙浓度([Ca2+]i),钙调蛋白量为指标,观察氯化镉(CdCl2,10-200μmolL-1)对脾细胞增殖的影响与细胞钙稳态的关系.结果表明,在本实验条件下,CdCl2对小鼠脾淋巴细胞转化,IL-2的产生有明显抑制作用,且与剂量和时间呈依赖关系.CdCl2对脾细胞钙稳态有干扰,致细胞[Ca2+]i浓度升高,钙调蛋白量下降,也与剂量和时间呈依赖关系.提示CdCl2对小鼠脾细胞增殖的抑制作用与细胞钙稳态失衡密切相关.
The effects of cadmium chloride (CdCl2, 10-200 μmol L-1) on the proliferation of mouse spleen lymphocytes, production of interleukin 2 (IL-2), intracellular free calcium concentration ([Ca 2+] i) Relationship between proliferation of spleen cells and calcium homeostasis. The results showed that under the experimental conditions, CdCl2 significantly inhibited the splenic lymphocyte transformation and IL-2 production in mice and was dose-dependent and time-dependent. CdCl2 interfered with the calcium homeostasis of splenocytes, resulting in an increase in the concentration of [Ca2 +] i in the cells and a decrease in calmodulin, also in a dose-dependent manner. Tip CdCl2 inhibition of mouse spleen cell proliferation and cell calcium imbalance is closely related.
