目的:考察化瘀通痹胶囊对风湿性关节炎的治疗作用。方法:给予大鼠Freund’s完全佐剂复制佐剂性关节炎模型。实验分为空白对照、模型对照(等容去离子水)、寒湿痹颗粒(3.5 g.kg-1)组与化瘀通痹胶囊高、中、低剂量(生药,1.1,0.55,0.27 g.kg-1)组,ig每天1次,连续14 d,实验期间于造模后第3,6,8,11,14,18,21,24天测量大鼠致炎足踝关节周长,第8,11,14,18,21,24天测量致炎对侧足踝关节周长,并于18,21 d测定致炎足机械性压迫所致的痛阈值;通过蛋清致大鼠足肿胀实验、二甲苯致小鼠耳肿胀实验、小鼠扭体实验,考察受试药物的抗炎、镇痛作用。实验分为空白对照、寒湿痹颗粒组与化瘀通痹胶囊高、中、低剂量组,大鼠给药同佐剂性关节炎实验,小鼠给药为空白对照、寒湿痹颗粒(4.5 g.kg-1)组与化瘀通痹胶囊高、中、低剂量(1.4,0.7,0.35 g.kg-1),每天1次,连续ig 7 d。结果:与模型组比较,化瘀通痹胶囊高、中剂量组均改善佐剂性关节炎模型大鼠致炎足18,21 d踝关节周长(P<0.05),高剂量组可显著提高致炎18,21 d大鼠机械痛阈值(P<0.05);中剂量组可显著提高致炎21 d大鼠机械痛阈值(P<0.05);与模型对照组比较,化瘀通痹胶囊高、中剂量组可抑制蛋清致炎后4h大鼠足跖肿胀度(P<0.05);高剂量组能降低二甲苯致小鼠耳肿胀的程度(P<0.05);高、中剂量组小鼠扭体反应次数明显减少(P<0.05)。结论:化瘀通痹胶囊具有良好的抗炎、镇痛作用,对风湿性关节炎具有明显的预防和治疗作用。
Objective: To investigate the therapeutic effect of Huayu Tongbi Capsule on rheumatoid arthritis. Methods: Freund’s complete adjuvant was used to replicate adjuvant arthritis model in rats. The experiment was divided into blank control, model control (isodose deionized water), cold and wet granules (3.5 g.kg-1) group and Huayu Tongbi capsule high, medium and low dose (crude drug, 1.1,0.55,0.27 g .kg-1) group, ig once a day for 14 days. The ankle joint circumference of rats was measured on the 3rd, 6th, 8th, 11th, 11th, 18th, On the 8th, 11th, 14th, 18th, 21st and 24th days, the periapical ankle joint circumference was measured, and the pain threshold caused by mechanical compression of the inflammatory foot was measured on 18 and 21 days. Experiments, xylene-induced mice ear swelling test, writhing mice experiments to examine the anti-inflammatory drugs, analgesic effect. The experiment was divided into blank control group, cold and wet group and Huayu Tongbi capsule high, medium and low dose group, the rats were given adjuvant arthritis experiment, the mice were given as blank control, 4.5 g.kg-1) and Huayu Tongbi Capsules at high, medium and low doses (1.4, 0.7, 0.35 g.kg-1) once daily for 7 days. Results: Compared with the model group, high and medium dose groups of Huayu Tongbi Capsule ameliorates the ankle joint circumference of 18 and 21 d (P <0.05) in the adjuvant arthritis model rats, and significantly increases in the high dose group (P <0.05). The middle dose group could significantly increase the mechanical pain threshold (P <0.05) of 21-day-induced inflammation in rats. Compared with the model control group, Huayu Tongbi capsule height (P <0.05); middle dose group could inhibit the degree of paw edema (P <0.05) in rats treated with albumen at 4h; high dose group could reduce the degree of ear edema caused by xylene in mice (P <0.05) Writhing reaction times decreased significantly (P <0.05). Conclusion: Huayu Tongbi Capsule has good anti-inflammatory and analgesic effects and has obvious preventive and therapeutic effects on rheumatoid arthritis.
