通心络对自发性糖尿病大鼠心肌组织胶原网络重构的影响

目的:观察通心络对自发性糖尿病(GK)大鼠心肌Ⅰ型和Ⅲ型胶原阳性表达及含量、细胞转化生长因子(TGF-β1)含量的影响。方法:4月龄GK大鼠30只,随机分为GK对照组、通心络大、小剂量组,另取4月龄Wistar大鼠10只作正常对照,分别灌胃12周。灌胃结束后,制备心肌切片,免疫组化染色,计算胶原I、胶原Ⅲ阳性面密度比值以代表表达程度;取心肌匀浆,酶联免疫方法测定心肌Ⅰ、Ⅲ型胶原及TGF-β1含量。结果:与同月龄正常大鼠相比较,GK大鼠血糖明显增高(P<0.01),通心络药物组有降低趋势,但无差异(P>0.05)。与正常大鼠比较,GK大鼠心肌胶原Ⅰ阳性面密度比值明显增高(P<0.01),胶原Ⅲ阳性面密度比值明显降低(P<0.01),心肌匀浆胶原Ⅰ含量明显增高(P<0.01),胶原Ⅲ含量降低(P<0.05),心肌胶原Ⅰ/Ⅲ比值明显增高(P<0.01),TGF-β1含量降低(P<0.01);与7月龄GK大鼠比较,通心络组胶原Ⅰ阳性面密度明显降低(P<0.01),高、低剂量组之间有差异(P<0.05),胶原Ⅲ阳性面密度明显升高(P<0.01);胶原Ⅰ含量明显降低(P<0.01),胶原Ⅲ含量明显提高(P<0.01),胶原Ⅰ/Ⅲ比值降低(P<0.01),TGF-β1含量明显提高(P<0.01),高、低剂量组之间无差异(P>0.05)。结论:通心络可降低GK大鼠心肌胶原Ⅰ阳性表达,Ⅰ型胶原含量,升高胶原Ⅲ阳性表达,Ⅲ型胶原含量,降低胶原Ⅰ/Ⅲ比值,升高TGF-β1含量,具有改善心肌间质网络重构作用。 OBJECTIVE: To observe the effect of Tongxinluo on the positive expression and content of type I collagen and type III collagen and the content of transforming growth factor (TGF-β1) in myocardium of spontaneously diabetic rats. Methods: Thirty 4-month-old GK rats were randomly divided into GK control group, Tongxinluo large-dose group and small-dose group. Compared with normal rats, the positive ratio of myocardial collagen I surface density in GK rats was significantly higher (P<0.01), the ratio of positive area ratio of collagen III was significantly lower (P<0.01), and the content of collagen I in myocardial homogenates was significantly higher (P<0.01). ), collagen III content decreased (P <0.05), myocardial collagen I / III ratio was significantly higher (P <0.01), TGF-β1 content decreased (P <0.01); compared with 7-month-old GK rats, Tongxinluo group The positive area density of collagen I was significantly decreased (P<0.01), there was a difference between high and low dose groups (P<0.05), and the positive area density of collagen III was significantly higher (P<0.01); collagen I content was significantly lower (P< 0.01), Collagen III content increased significantly (P <0.01), collagen I / III ratio decreased (P <0.01), TGF-β1 content increased significantly (P <0.01), there was no difference between high and low dose groups (P> 0.05). Conclusion: Tongxinluo can reduce the positive expression of collagen I, the content of type I collagen, increase the positive expression of collagen III, increase the content of type III collagen, decrease the ratio of collagen I/III, and increase the content of TGF-β1 in myocardium of GK rats.
