本试验以驱蚊草幼嫩茎段、茎尖、叶片为外植体,研究了不同灭菌时间对无菌系建立的影响,不同生长调节剂组合对芽增殖的影响,不同种类生长素的生根效果。结果表明:幼嫩茎段用 0.1%升汞灭菌7min接种到初代培养基上诱导效果最好,在MS+BA2.0mg/L+2,4-D0.2mg/L 上诱导率可达90%,且愈伤组织质量较好;丛生芽诱导的最佳培养基为MS+BA1.0 mg/L+2,4- D0.5mg/L;最佳生根培养基为1/2MS+NAA0.5mg/L。
In this experiment, the young stem segments, shoot tips and leaves of P. repens were used as explants to study the effects of different sterilization time on the establishment of asexual lines, the effects of different growth regulators on bud proliferation, the rooting effects of different kinds of auxin . The results showed that inoculation of primary stem with 0.1% mercuric chloride for 7min on the primary culture medium resulted in the best inducing effect. The induction rate could reach 90% on MS + BA2.0mg / L + 2,4-D0.2mg / L %, And the quality of callus is better. The best medium for inducing shoots is MS + BA1.0 mg / L and 2,4-D0.5 mg / L. The best rooting medium is1 / 2MS + NAA0. 5mg / L.
