目的研究岗梅根总皂苷的最佳提取工艺条件。方法采用Design Expert 8.0分析软件,在单因素的基础上运用Box-Behnken响应曲面设计法,以提取时间、乙醇浓度、溶剂用量和提取次数建立4因素3水平的实验模型,并确立提取回归方程;使用响应曲面法优化提取工艺。结果最佳的工艺条件为:提取时间100 min,乙醇体积分数为56.33%,溶媒用量为11倍,提取次数为2次。验证实验显示总皂苷平均转移率为71.92%,模型预测值的绝对误差小于3%。结论响应曲面法能合理地优化岗梅中总皂苷的提取工艺,可为岗梅的综合利用提供参考。
Objective To study the optimum extraction conditions of Gang Megan saponins. Methods Using Design Expert 8.0 software, the Box-Behnken response surface design method was used on the basis of single factor to establish a 4-level and 3-level experimental model with extraction time, ethanol concentration, solvent dosage and extraction times, and the extraction regression equation was established. Using Response Surface Method to Optimize Extraction Process. Results The optimum conditions were as follows: the extraction time was 100 min, the volume fraction of ethanol was 56.33%, the amount of solvent was 11 times and the number of extraction was 2 times. Validation experiments showed that the average transfer rate of total saponins was 71.92%, and the absolute error of model predictive value was less than 3%. Conclusion Response surface methodology can reasonably optimize the extraction process of total saponins in Gangmei, which can provide a reference for the comprehensive utilization of Gangmei.
