高等植物的线粒体基因组比较大,一般从200—2500kb,加上其组织结构相当复杂,研究起来比较困难.目前的研究途径主要是两条,一是借助于脉冲电泳技术进行的,如完整线粒体DNA的脉冲电泳分析、结合某些稀有酶切位点的限制性酶的酶切,某些线粒体大分子DNA的大尺度物理图谱的构建;二是构建线粒体基因组的物理图谱.但因为线粒体结构比较复杂,含有较多的重复序列,构建图谱的难度较大,而且构建的结果有待进一步的检验.现
