目的探讨孕鼠叶酸缺乏对幼鼠肺发育及其叶酸结合蛋白1基因(Folbp1)的影响。方法成熟雌性36只SD大鼠随机均分为A组(缺乏叶酸饲料的实验组)和B组(添加叶酸饲料的对照组),2周后与成熟SD雄性大鼠交配,生后7 d处死新生大鼠,留取肺组织标本。HE染色观察病理改变,实时荧光定量RT-PCR技术检测Folbp1 mRNA表达。结果与B组比较,A组肺组织结构紊乱,肺泡数量明显减少,肺间隔明显增宽,肺泡、孔腔形状较不规则,肺组织结构明显被破坏。A组肺组织中Folbp1 mRNA相对表达量低于B组(561.21±73.53 vs.1095.73±95.88)(P<0.05)。结论孕期叶酸缺乏会降低Folbp1基因表达,从而可能导致幼鼠肺发育形态异常,造成肺发育成熟障碍。
Objective To investigate the effects of folic acid deficiency on the development of lung and the expression of follistat binding protein 1 (Folbp1) in young rats. Methods Thirty-six female Sprague-Dawley rats were randomly divided into group A (experimental group lacking folate diet) and group B (control group fed with folic acid diet). Mice were mated with mature SD male rats 2 weeks later and sacrificed 7 days after birth Newborn rats, lung tissue samples were taken. Pathological changes were observed by HE staining and Folbp1 mRNA expression was detected by real-time fluorescence quantitative RT-PCR. Results Compared with group B, the structure of lung tissue in group A was disordered, the number of alveoli was obviously decreased, the interval between lungs was obviously widened, the shapes of alveoli and cavity were irregular, and the structure of lung tissue was obviously destroyed. The relative expression of Folbp1 mRNA in group A was lower than that in group B (561.21 ± 73.53 vs.1095.73 ± 95.88) (P <0.05). Conclusions Folic acid deficiency during pregnancy reduces Folbp1 gene expression, which may lead to abnormal lung development in young rats and cause obstructive pulmonary development.
