目的研究视黄醇(维生素A)对人脐带间充质干细胞表达表皮生长因子(EGF)、干细胞因子(SCF)、集落刺激因子1(CSF1)和白血病抑制因子(LIF)的影响。方法分离鉴定人脐带间充质干细胞,待细胞生长稳定后分为4组,分别用含12%FBS、12%FBS+1μmol/L视黄醇、15%血清替代品(KSR)和15%KSR+1μmol/L视黄醇的DMEM/F12培养液培养,3 d后收集细胞及培养液上清,用RT-PCR和ELISA分析视黄醇对人脐带间充质干细胞生成细胞因子EGF、SCF、CSF1和LIF的影响。结果分离所得细胞具备人脐带间充质干细胞免疫表型特征,细胞因子EGF、SCF和CSF1在m RNA和蛋白质水平均表达,而LIF只在m RNA水平表达;添加视黄醇(1μmol/L)后,SCF和CSF1在m RNA、蛋白质水平均显著增加(P<0.05),而EGF和LIF变化不显著(P≥0.05)。结论视黄醇(1 mol/L)可促进体外培养的人脐带间充质干细胞生成细胞因子SCF和CSF1。
Objective To investigate the effects of retinol on the expression of epidermal growth factor (EGF), stem cell factor (SCF), colony stimulating factor 1 (CSF1) and leukemia inhibitory factor (LIF) in human umbilical cord mesenchymal stem cells. Methods Human umbilical cord mesenchymal stem cells (MSCs) were isolated and identified. The cells were divided into four groups after they were stably cultured. The cells were stained with 12% FBS, 1% FBS and 1μmol / L retinol, 1μmol / L retinol in DMEM / F12 medium. After 3 days, the cells and culture supernatant were harvested, and the effects of retinol on the production of human umbilical cord mesenchymal stem cells (EGF), SCF and CSF1 And the impact of LIF. Results The isolated cells had the immunophenotypic characteristics of human umbilical cord mesenchymal stem cells. The expressions of cytokines EGF, SCF and CSF1 were expressed at m RNA and protein levels, while LIF mRNA was only expressed at m RNA level. Retinol (1 μmol / L) The levels of m RNA and protein in SCF and CSF1 were significantly increased (P <0.05), while the changes of EGF and LIF were not significant (P> 0.05). Conclusion Retinol (1 mol / L) can promote the production of cytokines SCF and CSF1 by human umbilical cord mesenchymal stem cells cultured in vitro.
