目的观察支气管哮喘模型小鼠外周血、支气管肺泡灌洗液(BALF)及肺组织中嗜酸性粒细胞(EOS)的改变以及佛手挥发油对以上指标的影响。方法C57小鼠以卵白蛋白(OVA)致敏造成哮喘模型,分为模型组,对照药物组,佛手挥发油高、中、低3个剂量组,另外10只为健康对照组。分别检测各组小鼠外周血、BALF中EOS的水平并观察肺组织病理切片中EOS的浸润情况。结果模型组外周血、BALF与对照组比较,EOS均明显增加(P<0.01);肺组织病理切片显示支气管及血管周围大量EOS浸润。各佛手挥发油组与模型组相比外周血BALF中EOS均有不同程度下降,高剂量组差异尤为显著(P<0.01)。各组肺组织病理切片中EOS浸润较模型组减少,剂量越大此作用越明显。结论佛手挥发油可抑制哮喘小鼠外周血、BALF中EOS水平,减少肺组织EOS浸润,拮抗气道炎症而发挥平喘作用,作用强度与剂量有关。
Objective To observe the changes of peripheral blood, bronchoalveolar lavage fluid (BALF) and eosinophil (EOS) in lung tissue of bronchial asthma model mice and the effects of volatile oil of bergamot on the above indexes. Methods C57 mice were sensitized with ovalbumin (OVA) to induce asthma model and divided into 3 groups: model group, control group and Bergamot high, medium and low dose groups. The other 10 were healthy control group. The levels of EOS in peripheral blood and BALF of mice in each group were detected and the infiltration of EOS in lung tissue sections was observed. Results Compared with the control group, the levels of EOS in the peripheral blood and BALF in the model group were significantly increased (P <0.01). The pathological sections of lung tissue showed a large number of EOS infiltration in the bronchus and blood vessels. Compared with model group, the EOS of BALF in each bergamot volatile oil group decreased to some extent, especially in high dose group (P <0.01). EOS infiltration in pathological sections of lung tissue in each group decreased compared with the model group, the greater the dose, the more obvious this effect. Conclusion Bergamot essential oil can inhibit EOS levels in peripheral blood and BALF of asthmatic mice, reduce EOS infiltration in lung tissue and antagonize airway inflammation and play an antiasthmatic effect. The intensity of action is dose-dependent.
