目的:通过观察福苓方对秀丽隐杆线虫寿命的影响,研究其延缓衰老的效果。方法:秀丽隐杆线虫分为空白组和福苓方高、中、低剂量组,通过寿命试验观察福苓方对线虫寿命影响,利用生殖力测试观察药物对线虫生殖力影响,急性热应激、氧化应激试验观察线虫在应激状态下寿命的改变,探讨该复方的作用机制。结果:各组的平均寿命分别为(19.96±1.48),(36.03±1.76),(28.47±1.01),(19.39±0.61)d,最大寿命依次为(21.01±1.36),(37.71±2.38),(29.19±3.12),(22.00±2.01)d。与空白组相比,福苓方高、中剂量组均能显著延长线虫的平均寿命、最大寿命及在氧化应激、热应激环境下寿命。福苓方组线虫总子代数目与空白组相比差异无统计学意义。结论:福苓方可延缓线虫衰老,其作用机制可能与该方能提高线虫的抗压力应激能力有关。
Objective: To observe the effect of Fuling on the life span of Caenorhabditis elegans, and to study its effect of delaying aging. Methods: Caenorhabditis elegans were divided into blank group and high, medium and low dose groups of Fufang Fang. The life span of Fuling was observed through life test. The effect of drug on nematode fertility was observed by fecundity test. Acute heat stress The oxidative stress test was used to observe the change of life expectancy of nematodes under stress and to explore the mechanism of action of this compound. RESULTS: The average life span of each group was (19.96 ± 1.48), (36.03 ± 1.76), (28.47 ± 1.01) and (19.39 ± 0.61) d, respectively, and the maximum life expectancy was (21.01 ± 1.36) and (37.71 ± 2.38) (29.19 ± 3.12), (22.00 ± 2.01) d. Compared with the blank group, high and medium dose groups of Fuling significantly prolonged the average life expectancy, maximum life expectancy and life expectancy of the nematodes under oxidative stress and heat stress. There was no significant difference in the number of nematodes in the Fuling group compared with the blank group. Conclusion: Fuling can delay nematode senescence and its mechanism may be related to its ability to enhance the anti-stress ability of nematodes.
