目的和方法 :测定双肾双夹 ( 2K2C)肾性高血压大鼠早期 ,左心室α -肌球蛋白重链 (α -MHC)和 β 肌球蛋白重链 ( β MHC)mRNA水平的变化。实验分四组 :对照组、2K2C组、巯甲丙脯酸组、巯甲丙脯酸对照组。术后 2 4h、36h、48h、72h测动脉血压 ,提取左心室总RNA ,各样本取 8.0 μg ,与 32 P标记的α MHC和 β -MHCcDNA探针作斑点杂交。结果 :高血压大鼠左心室α MHCmRA水平明显降低 ,β MHCLmRA水平明显升高 ,且该变化可明显被巯甲丙脯酸所抑制。结论 :肾性高血压早期 ,MHC基因表达发生改变 ,肾素—血管紧张素系统 (RAS)可能起重要作用
PURPOSE AND METHODS: The changes of α MHC and β MHC mRNA in the early stage of renal hypertensive rats with double renal clamp (2K2C) were determined. The experiment was divided into four groups: control group, 2K2C group, captopril group, captopril control group. Arterial blood pressure was measured at 24 hours, 36 hours, 48 hours and 72 hours after operation. Total RNA of left ventricle was extracted. Each sample was taken as a spot hybridization with 32 P-labeled α MHC and β-MHC cDNA probes. Results: The level of α MHCmRA in left ventricle was significantly decreased and the level of β MHCLmRA was significantly increased in hypertensive rats, and the change was obviously inhibited by captopril.
