脐带条件培养液(HUCCM)用于人骨髓细胞体外琼脂培养中,能生成典型的由粒系细胞组成的集落。其集落产率随着人脐带条件培养液剂量的增加而增加,以后趋于平坦;集落产率与培养时间有一定的关系。培养8天为最佳时间。此时,集落形态丰满,产率较高;集落的生成量与接种骨髓有核细胞数之间呈良好的线性关系。因而提供了一个定量的测试技术。人脐带条件培养液制备简单,活性高,个体间差异小,且脐带来源丰富,是一种较理想的集落刺激因子。
Umbilical cord conditioned medium (HUCCM) is used in in vitro agar culture of human bone marrow cells to produce typical colonies of myeloid cells. The yield of colony increased with the increase of dosage of human umbilical cord culture medium and became flat afterwards. The yield of colony had some relationship with the culture time. Cultivate 8 days is the best time. At this point, the colony was full and the yield was high. There was a good linear relationship between the colony formation and the number of nucleated cells inoculated with bone marrow. Thus providing a quantitative test technique. Human umbilical cord blood conditioned medium is simple, high activity, small differences between individuals, and rich in umbilical cord source, is an ideal colony stimulating factor.
