目的:研究联合应用布地奈德(BUD)和不对称二基精氨酸(ADMA)对哮喘大鼠气道炎症和Ⅲ型前胶元(PCⅢ)表达的影响,探讨该法预防哮喘气道重塑的意义。方法:SD大鼠40只,随机分为5组:A组(8只,哮喘模型组)、B组(8只,全量 ADMA处理哮喘组)、C组(8只,全量BUD处理哮喘组)、D组(8只,半量BUD+半量ADMA处理哮喘组)、E组(8只,正常对照组)。每周激发一次,12周后,处死大鼠,采集气道标本,制备石蜡切片,HE染色观察5组大鼠肺组织的病理变化,并用免疫组化二步法显示PCⅢ在气道的表达。结果:①A组大鼠可见气道大量炎症细胞浸润、上皮细胞脱落、血管增生、平滑肌层增厚及上皮下纤维化,这些变化在B组、C组和D组均有所减轻,其中C组和D组与E组比较差异不明显。②与E组比较,PCⅢ在A组大鼠的表达显著升高(P<0.001);C组和D组与E组比较差异无显著性(P>0.05)。结论:联合应用BUD和ADMA 可有效抑制哮喘大鼠气道炎症和PCⅢ表达,进而抑制气道重塑的发生。
OBJECTIVE: To study the effects of combination of budesonide (BUD) and asymmetric diyl arginine (ADMA) on the airway inflammation and the expression of procollagen III (PCⅢ) in asthmatic rats, Plastic meaning. Methods: Forty Sprague Dawley rats were randomly divided into 5 groups: group A (n = 8), group B (n = 8), group C (n = 8) , Group D (8, half amount of BUD + half ADMA treatment asthma group), group E (8, normal control group). The rats were sacrificed 12 weeks later. The airway specimens were collected and paraffin sections were made. The pathological changes of the lung tissue were observed by HE staining. The expression of PCⅢ in the airway was detected by immunohistochemical two-step method. Results: ①A group of rats showed a large number of airway inflammatory cell infiltration, epithelial cell shedding, vascular proliferation, thickening of the smooth muscle layer and subcutaneous fibrosis, these changes in groups B, C and D were relieved, of which C And D group and E group was not significantly different. ② Compared with E group, the expression of PCⅢ in group A was significantly increased (P <0.001). There was no significant difference between group C and group D and group E (P> 0.05). Conclusion: The combination of BUD and ADMA can effectively inhibit airway inflammation and PC Ⅲ expression in asthmatic rats, and then suppress airway remodeling.
