应用人大肠癌细胞裸鼠脾移植肝转移模型研究大肠癌HT29、Lovo细胞系转移能力的差异,然后应用漂白后荧光恢复技术,使用粘附式细胞仪对不同转移能力的HT29、Lovo细胞的缝隙连接细胞间通信功能进行检测。结果表明,HT29细胞肝转移率为4/8,Lovo细胞肝转移率为8/8。HT29细胞的荧光恢复百分率较高,为(39.26±16.94)%,明显高于Lovo细胞(19.67±14.42)%。说明大肠癌细胞转移能力的增加可能与其缝隙连接细胞间通信功能的丧失有关
Human liver cancer metastasis model of human colorectal cancer cells in nude mice was used to study the difference in the metastatic potential of colorectal cancer HT29 and Lovo cell lines. Then the fluorescence recovery after bleaching was applied. The gaps between HT29 and Lovo cells with different metastatic potentials were examined using an adherent cytometer. Connect the cell-to-cell communication function for detection. The results showed that the liver metastasis rate of HT29 cells was 4/8, and the liver metastasis rate of Lovo cells was 8/8. The percentage of fluorescence recovery of HT29 cells was higher (39.26±16.94)%, which was significantly higher than that of Lovo cells (19.67±14.42)%. Explains that the increased metastatic capacity of colorectal cancer cells may be related to loss of communication between gap junction cells
