研究了载有抗癌药物5-氟脲嘧啶(5-Fu)的磁性牛血清白蛋白微球(FMAM)的药物体外释放测定条件以及与释放相关的特性。实验表明测定的灵敏度和准确度与试液的pH相关,实验提示,在pH6.05~7.50区间内5-Fu可发生烯醇型互变异物转化,用紫外分光法可以测定转化率,实验结果说明微球制备固化温度愈高,则药物释放速度愈慢,在相同的制作工艺下,磁性流体投量增加,则释放速度加快,但在该实验条件下,磁性流体增加至50ul以上时,不受影响.
The in vitro release assay conditions and release-related properties of magnetic bovine serum albumin microspheres (FMAM) loaded with the anticancer drug 5-fluorouracil (5-Fu) were studied. Experiments show that the sensitivity and accuracy of the assay are related to the pH of the test solution. Experiments show that the conversion of enol-type tautomers can occur in 5-Fu within the range of pH6.05-7.50, and the conversion can be measured by UV spectrophotometry. The experimental results show that the higher the curing temperature, the slower the drug release rate. Under the same manufacturing process, the increase of magnetic fluid dosage increases the release rate, but under the experimental conditions, when the magnetic fluid is increased to more than 50ul , Not affected.
