目的:探讨慢性间歇性低压低氧(CIIH)预处理对大鼠脑低氧损伤后顶叶皮层c-Fos、Bcl-2表达的影响。方法:雄性成年大鼠48只,随机分为4组:正常对照组(CON)、慢性间歇性低压低氧组(CIIH)、急性低氧组(AH)和低氧预处理组(CIIH+AH)。采用甲苯胺蓝染色、免疫组织化学、透射电镜等方法观察大鼠脑顶叶皮层c-Fos、Bcl-2表达的变化。结果:CIIH组未见损伤变性的改变,与CON组相比差异无显著性(P>0.05)。与CON组和CIIH组相比较,AH组可见大鼠脑顶叶皮层神经元出现较为严重的损伤变性的改变,c-Fos、Bcl-2表达显著增加(P<0.01)。与AH组相比,CIIH+AH组神经元损伤变性减轻,Bcl-2表达增高(P<0.05),c-Fos表达降低(P<0.05)。结论:CIIH预处理可能通过上调大鼠脑低氧损伤后Bcl-2表达,下调c-Fos表达,抑制缺氧对神经元凋亡的诱导作用,从而发挥神经保护作用。
Objective: To investigate the effect of chronic intermittent hypobaric hypoxia (CIIH) preconditioning on the expression of c-Fos and Bcl-2 in rat parietal cortex after hypoxic injury. Methods: Forty-eight male adult rats were randomly divided into 4 groups: normal control group (CON), chronic intermittent hypobaric hypoxia group (CIIH), acute hypoxia group (AH) and hypoxic preconditioning group ). The changes of c-Fos and Bcl-2 expression in the parietal cortex were observed by toluidine blue staining, immunohistochemistry and transmission electron microscopy. Results: There was no change in lesion degeneration in CIIH group, but no significant difference compared with CON group (P> 0.05). Compared with CON group and CIIH group, the expression of c-Fos and Bcl-2 in AH group was significantly increased (P <0.01). Compared with AH group, the degeneration of neuronal injury, Bcl-2 expression increased (P <0.05) and c-Fos expression decreased (P <0.05) in CIIH + AH group. CONCLUSION: CIIH preconditioning may play a neuroprotective role by up-regulating the expression of Bcl-2 and down-regulating the expression of c-Fos after hypoxic injury in rat brain and inhibiting the induction of neuronal apoptosis by hypoxia.
