目的研究肝部分切除术对人结肠癌Caco-2细胞肝转移的影响及其相关机制。方法应用人结肠癌Caco-2细胞系,构建结肠癌肝转移裸小鼠模型,同时行肝部分切除术(PH),假手术(Sham)设为对照,并对Caco-2细胞行siRNA-Snail干扰。观察肝转移灶面积,肿瘤组织Snail和基质金属蛋白酶2蛋白(MMP-2)表达。结果 PH组转移灶面积、Snail蛋白和MMP-2表达均显著大于Sham组和siRNA-Snail-PH组。结论 PH可促进结肠癌发生肝转移,该作用与促进Snail表达及上调Snail调控的MMP-2表达相关。
Objective To investigate the effect of partial hepatectomy on hepatic metastasis of human colon carcinoma Caco-2 cells and its related mechanism. Methods Human colon cancer Caco-2 cell line was used to establish a model of hepatic metastasis in colon cancer. At the same time, partial hepatectomy (PH) and sham operation (Sham) were used as control. Caco-2 cells were treated with siRNA- interference. The area of liver metastasis, the expression of Snail and MMP-2 in tumor tissue were observed. Results The area of metastasis, the expression of Snail protein and MMP-2 in PH group were significantly higher than those in Sham group and siRNA-Snail-PH group. Conclusion PH can promote the hepatic metastasis of colon cancer, which is related to the promotion of Snail expression and the up-regulation of Snail-regulated MMP-2 expression.
