甲型肝炎病毒(HAV)的免疫测定是利用细胞培养获得的HAV抗原检测HAV特异性抗体。目前,生产HAV抗原的方法是组织培养,不但耗时而且价格昂贵。作者先前已经在重组痘苗病毒中表达了HAV的开放阅读框架(rV-ORF)。重组HAV多蛋白被精确地加工并装配成亚病毒颗粒。这种颗粒可与HAV中和抗体结合并能激发可被市售免疫测定试剂盒检出的抗体。本研究是将传统的细胞接种法生产的HAV抗原与重组痘苗病毒系统表达的HAV抗原作一比较。另外,还对HAV与rV-ORF抗原在免疫诊断测定中的应用进行了评价。
An immunoassay for Hepatitis A virus (HAV) detects HAV-specific antibodies using HAV antigen obtained from cell culture. At present, the method of producing HAV antigen is tissue culture, which is not only time-consuming but also expensive. The authors have previously expressed the open reading frame (rV-ORF) of HAV in recombinant vaccinia virus. Recombinant HAV polyprotein is precisely engineered and assembled into subviral particles. Such particles can bind to HAV neutralizing antibodies and can excite antibodies detectable by commercially available immunoassay kits. In this study, the HAV antigen produced by the conventional cell inoculation method was compared with the HAV antigen expressed by the recombinant vaccinia virus system. In addition, the applications of HAV and rV-ORF antigens in immunodiagnostic assays were also evaluated.
