本文应用TEM技术、电镜细胞化学、胶体金标记和银染技术结合研究了大鼠(Ratusnorvegicusvar.albulus)骨髓细胞染色体骨架。超薄切片结合RNP优先染色法在染色体原位显示RNP,同时又结合银染电镜技术及RNase-胶体金定位显示RNA,结果显示金颗粒与银颗粒在染色体中分布的位置与RNP所在部位恰好一致。说明RNP中的蛋白质含有银染的酸性蛋白,即所观察到的染色体骨架非组蛋白、RNP中的蛋白部分、银染蛋白三者是一致的。
In this paper, the chromosomal framework of bone marrow cells of rat (Ratus norvegicus var. Albulus) was studied by TEM, electron microscopy cytochemistry, colloidal gold labeling and silver staining. The ultrathin sections combined with RNP-preferred staining showed RNP in situ on the chromosome, combined with silver staining and RNase-colloidal gold localization showed that the distribution of gold particles and silver particles in the chromosome was exactly the same as that of RNP . This shows that the proteins in RNP contain silver-stained acidic proteins, that is, the observed chromosomal framework non-histone, the protein part in RNP, and the silver stained protein are consistent.
