1 The origin of biochemical regulation (Biochemical Modulation BCM) Biochemical regulation has been used in clinical cancer therapy for more than 15 years. In 1977, Yale University Bertino et al. used Sarcoma 180 as a cancer mouse test to observe the dependence of MTX/5-FU administration sequence. Using the survival rate as an indicator, first give MTX, and then give 5-FU after 2 hours, and get a multiplier effect. On the contrary, if 5F-U or two drugs are administered at the same time, the above results cannot be obtained. They believe that the main mechanism of action is the formation of a ternary complex of formyltetrahydrofolate (mTHF), fluorodeoxyuridine monophosphate (FdUMP) and thymidylate synthase (TS), which inhibits TS activity.
