目的观察不同溶液体系中三七总皂苷的分子聚集缔合特点,及β-环糊精对分子缔合的影响。方法光度法测定总皂苷-环糊精的包合常数Ka;总皂苷浓度Ci的特征吸收Ai,以摩尔吸收系数εi与Ci关系考察溶液体系总皂苷的分子缔合行为,并测定缔合度。结果三七总皂苷与β-环糊精能形成稳定包合物(Ka为464.3 L·mol-1);纯水、人工胃液、人工肠液体系中,总皂苷纯品无明显分子缔合,而粗提液中总皂苷在低浓度下即发生4分子缔合,对存在缔合现象的体系加入β-环糊精,总皂苷的分子缔合现象明显减弱。结论β-环糊精对总皂苷的包合作用减弱了三七粗提液中总皂苷的分子缔合现象。
Objective To observe the molecular aggregation and association characteristics of Panax notoginseng saponins in different solution systems and the influence of β-cyclodextrin on the molecular association. Methods The photometric method was used to determine the inclusion constants of total saponin-cyclodextrin, Ka, the characteristic absorption of total saponin Ci, and the molecular association behavior of the total saponins of the solution system was investigated by the relationship between the molar absorption coefficient εi and Ci. Results The total saponins of Panax notoginseng and β-cyclodextrin could form a stable inclusion complex (Ka was 464.3 L · mol-1). In pure water, artificial gastric juice and artificial intestinal juice, the total saponins had no obvious molecular association, Total saponin in the crude extract at a low concentration that occurs 4 molecules associate, there is an association phenomenon of the system by adding β-cyclodextrin, total saponin molecular association significantly weakened. Conclusion The inclusion of total saponins by β-cyclodextrin weakens the molecular association of total saponins in the crude extract of Panax notoginseng.
