目的:研究红花多糖的提取纯化工艺,确立红花多糖的最佳提取条件。方法:用水提醇沉法提取红花多糖,经过Sevage法除蛋白质,H2O2脱色,无水乙醇、丙酮、乙醚洗涤。通过对不同的提取次数、液料比及提取时间等实验,以及对粗多糖纯化的研究,确立各因素条件,采用苯酚—硫酸法测定其多糖含量。结果:各因素条件为提取时间1 h,液料比30∶1,提取4次,用Sevage、无水乙醇、丙酮、乙醚纯化。由此得红花中粗多糖的得率为6.5%,纯化后的多糖得率为4.7%。测定提取的粗多糖中多糖含量为70.315%,纯化后的多糖含量为76.048%。结论:方法纯化效果好,测定重现性好,而且成本低,操作简便快捷,适合工业生产。
OBJECTIVE: To study the extraction and purification of safflower polysaccharides and establish the optimal extraction conditions for safflower polysaccharides. Methods: The safflower polysaccharide was extracted by water extraction and alcohol precipitation. The protein was removed by Sevage method, and bleached by H2O2, washed with absolute ethanol, acetone and ether. Through experiments on different extraction times, liquid-to-solid ratio and extraction time, as well as the research on purification of crude polysaccharides, the factors and conditions were established. The polysaccharide content was determined by phenol-sulfuric acid method. Results: The conditions were as follows: extraction time 1 h, liquid to liquid ratio 30:1, extraction 4 times, purification with Sevage, absolute ethanol, acetone and ether. The yield of crude polysaccharide in safflower was 6.5% and the yield of purified polysaccharide was 4.7%. The content of polysaccharide in crude polysaccharide was 70.315% and the content of purified polysaccharide was 76.048%. Conclusion: The method has good purification effect, good reproducibility, low cost, simple and quick operation, suitable for industrial production.
